Abstract :The interaction of keyhole limpet hemocyanin ( KL H ) and PEG2002nip hensamide was 
investigated by fluorescence spectroscopy , ultraviolet absorption spectroscopy and circular dichroism(CD) spectroscopy. It was shown that KL H fluorescence at 340 nm was quenched regularly wit h theaddition of PEG2002nip hensamide ; t he quenching constant s were decreased with the temperatureincreasing ; and the quenching was verified as static fluorescence quenching. The binding constant s ( K)were obtained according to Lineweaver2Burk equation at different temperature. The calculatedthermodynamic parameter s ( △H , △S) according to Van’t Hoff equation indicated t hat the elect rostaticinteraction played major roles in t he binding processes. The distance between KL H and PEG2002nip hensamide was 5. 76 nm according to t he theory of the FÊrster energy t ransference. The effect s ofPEG2002nip hensamide on the conformation of KL H were f urt her analyzed by synchronous fluorescencespectroscopy and circular dichroismspectroscopy. PEG2002nip hensamide could be deposited andtransported by KL H and it affected t he conformation of KL H. The secondary structure of KL H wasaltered (CD data) , i. e. , t heα2helices were reduced f rom 43. 1 % to 37. 8 %.
